Loop-mediated isothermal amplification (LAMP) is a new nucleic acid amplification method. To be specific, 4 specific primers are designed for 6 sections of the target gene; under the action of Bst DNA polymerase, isothermal amplification is performed in a condition of 60-65℃; in this way, ×10^9～10^10 nucleic acid amplification can be realized in 15-60 minutes. Clearly, such a method is featured with ease of operation, strong specificity and ease of product detection, etc.. During DNA synthesis, pyrophosphate ions separated out from dNTPs react with magnesium ions in the reaction solution, which leads to lots of magnesium pyrophosphate sediments that are white. Therefore, turbidity can be used as a reaction index; and, amplification can be identified by observing turbid white sediments by naked eyes, which does not require electrophoresis or ultraviolet visualization. Under the circumstance that loop-mediated isothermal amplification based reactions can be fulfilled with no need for PCR instrument or expensive reagents, it has a broad application prospect.

As a new nucleic acid amplification technique, LAMP has been widely applied in the field of molecular biological detection. In comparison with other existing nucleic acid amplification techniques, LAMP has many unique advantages:
1、Based on LAMP, amplification can be realized under isothermal conditions without initial denaturation of the template, which not only reduces the influence incurred by temperature rise and fall required by PCR, but also lowers the requirement for expensive and precision experimental instrument. In addition, corresponding amplification efficiency is also improved. For example, several copied destination sequences can be rapidly amplified to 10^10 copies in 1 hour.
2、Respectively, a pair of outer primers and a pair of inner primers are respectively selected based on the LAMP technology to identify 6 different sections on the destination sequence. Moreover, it is highly selective to the destination sequence. This lowers the influence of non-target sequences. Therefore, specificity of amplification is significantly strong.
3、Positive amplification reaction may result in massive products with a structure similar to cauliflower and white pyrophosphoric acid sediments. In order to test amplification products, there are many available methods, including gel electrophoresis and visual inspection; alternatively, dyes may be added into a reaction system to complete detection according to color change; or, a turbidity meter can be used to perform real-time quantitative analysis on primitive nucleic acid molecules according to differences in turbidity of amplification products. Additionally, simply treated samples can be directly adopted as templates for some pathogene, which gets rid of a tedious DNA/RNA extraction procedure. To sum up, LAMP, being rapid, efficient, specific, sensitive and economical, has been applied in detection of pathogenic bacteria, parasites, viruses, diseases and transgenic product. In the future, it may be also widely used in fields of clinical diagnosis, environmental monitoring and food source safety, etc.. In other words, LAMP possesses an enormously broad prospect of development and application.
